Email Reader's privacy can be revealed if you send something as email say a greeting card or some advertisement with some image which it fetches from your domain you have registered
or sending a link to reader
If reader clicks the link
And page loads from your site
you get IPAddress and get location
But dont rely on this
If the reader is using HTTP proxy sites
or is behind some proxy caching server configured in network which doesnot reveal internal network IP Addresses
Detailed EMail Headers also reveal the path through which email reached your mail server
But not so reliable
So nothing can be said for sure
